Özet

In this study, novel peppermint oil-based organohydrogel (PORH) electrodes were developed for the electrochemical detection of CA-125 and CA 15-3 cancer biomarkers in serum and blood samples. The PORHs were synthesized using agar, glycerol, peppermint oil, and methylene bisacrylamide (MBA) or glutaraldehyde (GA) as cross-linking agents. The incorporation of peppermint oil improved the structural stability and electrochemical activity of the organohydrogel system due to its multifunctional natural components. The synthesized PORHs were characterized by Fourier transform infrared (FT-IR) spectroscopy, scanning electron microscopy (SEM), and swelling tests. The electrochemical performance of the fabricated electrodes was investigated using cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and differential pulse voltammetry (DPV). Among the synthesized electrodes, PORH-5 exhibited the highest electrochemical response, showing current density of 1.45 mA/cm2 for CA-125 (1000 ng/mL) and 1.12 mA/cm2 for CA 15-3 (500 ng/mL). The developed sensor demonstrated linear detection ranges of 1–100 ng/mL for CA-125 and 0.5–150 ng/mL for CA 15-3. The limits of detection (LOD) were calculated as 0.38 µU/mL for CA-125 and 0.52 µU/mL for CA 15-3 (S/N = 3). In addition, interference studies confirmed the high selectivity of the PORH-5 electrode against common serum components. The results demonstrate that the peppermint oil-based PORH platform provides a simple, biocompatible, and sensitive electrochemical sensing system for the detection of cancer biomarkers without requiring complex nanomaterials or biological recognition elements.
