Geleneksel Yerel Beyaz Peynir Örneklerinden İzole Edilen Laktik Asit Bakterilerinin MALDI-TOF MS Yöntemi ile Tanımlanması ve Antibiyotik Direnç Profillerinin Belirlenmesi

Özet

Bu çalışmada, Eskişehir’de üretilen geleneksel salamura peynir örneklerinden izole edilen Laktik Asit Bakterilerinin (LAB) antibiyotik direnç profili incelenmiştir. Peynir üretiminde uygulanan tekniksel çeşitlilik ve ürün tiplerindeki zenginlik, Eskişehir’in bu araştırma için örneklem alanı olarak seçilmesinde temel etken olmuştur. Peynirlerin mikrobiyotası kullanılan süt kaynakları, çiğ süt işleme koşulları ve spesifik fermantasyon uygulamaları gibi yerel faktörlerden büyük ölçüde etkilenmektedir. Farklı orijin ve işleme yöntemlerinden elde edilen toplam on iki temsili peynir örneği analiz edilmiştir. Etkinliği ve hızlı sonuç vermesi nedeniyle LAB düzeyinde tür tanımlaması için MALDI-TOF MS kullanılmıştır. Tanımlanan 25 izolatın %48’ini Enterococcus faecium, %28’ini Lactobacillus plantarum, %12’sini Enterococcus durans, %8’ini Lactococcus lactis ve %4’ünü Lactococcus garvieae oluşturdu.Antibiyotik duyarlılık testi Klinik Laboratuvar Standartları Enstitüsü (CLSI) yönergelerine göre Kirby-Bauer disk difüzyon yöntemi ile yapıldı. Klindamisin (%84), eritromisin (%76) ve rifampisin (%72) için yüksek direnç oranları kaydedilirken, tetrasiklin ve kloramfenikol için hiçbir direnç kaydedilmedi. Bu bulgular, LAB’lerinin direnç genlerinin rezervuarları olarak rolünü ve patojenik bakterilere potansiyel yatay gen aktarımı nedeniyle halk sağlığı riski oluşturduğunu vurgulamaktadır. Çalışma, geleneksel peynirlerde sıkı güvenlik değerlendirmelerine ve hayvancılıkta antibiyotik kullanımına ilişkin düzenleyici mevzuata duyulan ihtiyacın altını çizmektedir. LAB’lerinin daha fazla direnç mekanizması ve genetik çalışması önerilmekte olup, gıda güvenliği üzerinde önemli bir etkiye sahiptir.
This study investigated the antibiotic resistance profiles of Lactic Acid Bacteria (LAB) isolated from traditional brined cheeses produced in Eskişehir, Turkey. The diversity of cheese production techniques and the richness of product types were key factors in selecting Eskişehir as the sampling area for this study. The microbiota of cheeses is significantly influenced by local factors such as milk sources, raw milk processing conditions, and specific fermentation practices. A total of twelve representative cheese samples from different origins and processing methods were analyzed. MALDI-TOF MS was employed for species identification at the LAB level due to its effectiveness and speed. Among the 25 identified isolates, Enterococcus faecium represented 48%, Lactobacillus plantarum 28%, Enterococcus durans 12%, Lactococcus lactis 8%, and Lactococcus garvieae 4%. The antibiotic susceptibility test was performed by the Kirby-Bauer disk diffusion method according to Clinical Laboratory Standards Institute (CLSI) guidelines. High resistance rates were recorded for clindamycin (84%), erythromycin (76%), and rifampicin (72%), while no resistance was recorded against tetracycline and chloramphenicol. These findings highlight the role of LAB as reservoirs of resistance genes, posing a public health risk due to potential horizontal gene transfer to pathogenic bacteria. The study underlines the need for stricter safety assessments in traditional cheeses and regulatory legislation on the use of antibiotics in livestock. Further resistance mechanisms of LAB and their genetic study are recommended, having an important bearing on food safety.
